What is the main steps of whole transcriptome sequencing?
The samples of whole transcriptome sequencing are constructed to two libraries, one small RNA library (including miRNA) and one ribosome removal strand specific library (including mRNA, lncRNA, and circRNA), and then two libraries are sequenced, respectively. Finally, the sequence information of four kinds of RNA -- miRNA, lncRNA, mRNA, circRNA are obtained.
Why do researchers study the whole transcriptome?
There are different kinds of RNA in organisms. These RNA molecules are all related and interacted with each other. Single mRNA or ncRNA research has been unable to meet the needs of scientific research. Whole transcriptome has become a powerful tool to explain biological problems by combining multiple RNA information for integrated analysis and exploring the potential regulatory network mechanism!
