Which species can study lncRNA?
For lncRNA analysis, there are the following requirements for species.
a. Species are eukaryotes.
b. Species have reference genome.
c. There are complete annotations.
Why does lncRNA sequencing need to construct the strand specific library?
lncRNA Sequencing data can determine whether the transcripts are from sense or antisense DNA chains. Compared with ordinary transcriptome sequencing, it can more accurately count the number of transcripts and identify the structure of genes. At the same time, it can find more antisense transcripts and better identify antisense lncRNA.
Why is it necessary to remove rRNA when lncRNA sequencing and library construction?
In lncRNA, only the 3 'end of lincRNA has polyA structure, and other lncRNAs do not have polyA structure. Therefore, in order to obtain more complete lncRNA information, it is not recommended to use the way of oligo (dT) magnetic bead enrichment method.
What is lncRNA sequencing?
Long noncoding RNA (LncRNA) is a class of RNA molecules that are more than 200nt. They rarely have protein encoding potentials, but they can bind DNA, RNA and protein. They regulate the expression level of genes at various levels (epigenetic regulation, transcriptional regulation, post transcriptional regulation) in the form of RNA. lncRNA sequencing is to study all the lncRNA and mRNA transcribed from specific tissues or cells of existing reference genome species in a specific period.
