  • Why does lncRNA sequencing need to construct the strand specific library?


     lncRNA Sequencing data can determine whether the transcripts are from sense or antisense DNA chains. Compared with ordinary transcriptome sequencing, it can more accurately count the number of transcripts and identify the structure of genes. At the same time, it can find more antisense transcripts and better identify antisense lncRNA.


  • Why is it necessary to remove rRNA when lncRNA sequencing and library construction?


     In lncRNA, only the 3 'end of lincRNA has polyA structure, and other lncRNAs do not have polyA structure. Therefore, in order to obtain more complete lncRNA information, it is not recommended to use the way of oligo (dT) magnetic bead enrichment method.
